Default Working on large css files

I have a large css file that I want to modify (about 850 lines) and I am wondering what would be the best way to do that.
I've been looking at Chrome's Developer Tools and I am able to find code related to particular parts of the website and turn it on and off to see what it does etc (checkboxes).
I've been reading a lot about css and html on the net lately, and I have experience also, but redesigning a large website like this (video tube script) does not seem like an easy thing to do etc.

I was wondering if somebody could tell me, from experience, what would be the best way to approach that and how not to spend 3 years figuring out what is what in the code and how it relates to other parts etc.

Thanks in advance for any info.

I think the Software developers and expertise can help you out a lot about it... You can also check your query by goggling it on the internet... I am sure that you would find a lot of relevant topics related to this.

It will depend on what you want to modify, I use simple programs like allycode and notepad++ to play about with CSS, alleycode is good because you can have a high resolution and you see all the code.

Give us an idea what you are going to do, things like simple label changes for seo, adding totally new content, or just optimising the CSS.

I want to redesign a large video tube script website. I am still preparing myself to do this by learning php (and Smarty - thats what it uses) as well as JavaScrtipt. I want to know exactly what is what and understand entire code before I start. There is 126 templates that fall together in different ways depending on what a user selects on the screen etc. I think I can get rid of at least half of them right from the start though (features that I am not gonna be using etc).
I am sure it will be a while before I start, but the major part will be the CSS for sure
    Thanks.

EDIT: I want to change how it looks (color scheme, fonts, window sizes, positions of particular elements etc)

I want to make it look attractive (some websites just look good if you know what I mean, not necessarily adult), it is possible, but it dont seem to be easy to achieve.
